ONE SIMPLE APPROACH IS TO ALWAYS MAKE SURE THE IMAGE IS IN A PREDICTABLE STATE BY REDRAWING IT:

public void mouseMove(int x,int y)  {
    pen.drawImage(mapImage); 
    if (y > 65) {
       pen.invertRect(0,65,220,65);
    }  
    if (y <= 65) {
       pen.invertRect(0,0,220,65);
    }	
}

THIS VERSION REDRAWS THE IMAGE MUCH MORE FREQUENTLY THAN NECESSARY.
